    Seed particle formation for silicate dust condensation by SiO nucleation

    Clustering of the abundant SiO molecules has been discussed as a possible mechanism of seed particle formation for silicate dust in stellar outflows with an oxygen rich element mixture. Previous results indicated that condensation temperatures based on this mechanism are significant lower than what is really observed. This negative result strongly rests on experimental data on vapour pressure of SiO. New determinations show the older data to be seriously in error. Here we aim to check with improved data the possibility that SiO nucleation triggers the cosmic silicate dust formation. First we present results of our measurements of vapour pressure of solid SiO. Second, we use the improved vapour pressure data to re-calibrate existing experimental data on SiO nucleation from the literature. Third, we use the re-calibrated data on SiO nucleation in a simple model for dust-driven winds to determine the condensation temperature of silicate in stellar outflows from AGB stars. We show that onset of nucleation under circumstellar conditions commences at higher temperature than was previously found. Calculated condensation temperatures are still by about 100 K lower than observed ones, but this may be due to the greenhouse effect of silicate dust temperatures. The assumption that the onset of silicate dust formation in late-type M stars is triggered by cluster formation of SiO is compatible with dust condensation temperatures derived from IR observations.Comment: 11 pages, 11 figure

    Mobile Phone Data for Mapping Urban Dynamics

    During the last few years, new tools for estimating people’s density in cities have emerged through mobile network data. As opposed to the more traditional methods of urban surveys, the use of aggregated and anonymous mobile phone network log files has shown promise for large-scale surveys with notably smaller efforts and costs. Moreover, a frequent data feed from the mobile network has been argued to demonstrate fine grain over-time variation in urban movements, lacking from the traditional prediction methods. Despite the positivist approach to the new methodology, additional evidence is needed to show how mobile network data correlate with the actual presence of people, and how they can be used to map different urban domains. We try to address this shortcoming presenting the results of a research carried out in Lombardy Region, using mobile phone data provided by Telecom Italia, as a promising approach to assist the traditional database and analysis of urban dynamics as new challenges for urban plannin

    Deamidation at Asparagine and Glutamine As a Major Modification upon Deterioration/Aging of Proteinaceous Binders in MuralPaintings

    Proteomic strategies are herein proved to be a complementary approach to the well established amino acid composition analysis for the characterization of the aging and deterioration phenomena occurring to proteinaceous materials in works-of-art. Amino acid analyses on several samples demonstrated that proteins in the frescoes from the Camposanto Monumentale in Pisa are deteriorated as revealed by the decrease in Met, Lys, and Tyr content and by the presence in all the samples of amino malonic acid as a result of Ser, Phe, and Cys oxidation. Proteomic analysis identified deamidation at Asn and Gln as a further major event occurred. This work paves the way to the exploitation of proteomic strategies for the investigation of the molecular effects of aging and deterioration in historical objects. Results show that proteomic searches for deamidation by liquid chromatography-tandem mass spectrometry (LC-MS/MS) could constitute a routine analysis for paintings or any artistic and historic objects where proteins are present.     The cellular mechanism(s) underlying autism spectrum disorders (ASDs) are not fully understood although it has been shown that various genetic and environmental factors contribute to their etiology. As increasing evidence indicates that astrocytes and microglial cells play a major role in synapse maturation and function, and there is evidence of deficits in glial cell functions in ASDs, one current hypothesis is that glial dysfunctions directly contribute to their pathophysiology.     Preliminary slip rate estimates for the Düzce segment of the North Anatolian Fault Zone from offset geomorphic markers

    New estimates on the Quaternary slip rate of the active transform margin of North Anatolia are provided. We investigated the area struck by the 12th November 1999, Mw 7.1 earthquake, that ruptured the Düzce fault segment of the North Anatolian Fault. In order to analyze the spectacular tectonically driven cumulative landforms and the drainage pattern settings, we carried out a 1:25,000-scale geological and geomorphological mapping along the fault trace. We reconstruct and describe, as offset geomorphic markers, right-hand stream deflections and fluvial terraces inset into alluvial fan deposits. Radiocarbon dating indicates that ~100 m stream deflections were built up by the last ~7000 yrs of fault activity. Conversely, two documented and correlated Late Pleistocene fluvial terraces are horizontally offset by ~300 and ~900 m, respectively. These were dated by means of Optically Stimulated Luminescence (OSL) to ~21 ka BP and 60 ka BP. Assuming a constant rate of deformation for the Düzce Fault, ages and related offsets translate to consistent slip-rates that yield an average slip-rate of 15.0 ± 3.2 mm/yr for the last 60 ka. Thus, the Düzce Fault importantly contributes to the North Anatolian margin deformation, suggesting a present-day partitioning of displacement rates with the Mudurnu fault to the south and confirming its important role in the seismic hazard of the area

    Analysis of synaptic-like microvesicle exocytosis of B-cells using a live imaging technique.

    Pancreatic β-cells play central roles in blood glucose homeostasis. Beside insulin, these cells release neurotransmitters and other signaling molecules stored in synaptic-like microvesicles (SLMVs). We monitored SLMV exocytosis by transfecting a synaptophysin-pHluorin construct and by visualizing the cells by Total Internal Reflection Fluorescence (TIRF) microscopy. SLMV fusion was elicited by 20 mM glucose and by depolarizing K(+) concentrations with kinetics comparable to insulin secretion. SLMV exocytosis was prevented by Tetanus and Botulinum-C neurotoxins indicating that the fusion machinery of these organelles includes VAMP-2/-3 and Syntaxin-1, respectively. Sequential visualization of SLMVs by TIRF and epifluorescence microscopy showed that after fusion the vesicle components are rapidly internalized and the organelles re-acidified. Analysis of single fusion episodes revealed the existence of two categories of events. While under basal conditions transient fusion events prevailed, long-lasting episodes were more frequent upon secretagogue exposure. Our observations unveiled similarities between the mechanism of exocytosis of insulin granules and SLMVs. Thus, diabetic conditions characterized by defective insulin secretion are most probably associated also with inappropriate release of molecules stored in SLMVs. The assessment of the contribution of SLMV exocytosis to the manifestation of the disease will be facilitated by the use of the imaging approach described in this study

    A combination of spectroscopic techniques, hydrogen/deuterium exchange, and limited proteolysis experiments coupled to mass spectrometry analysis was used to depict the topology of the monomeric M*partly folded intermediate of aspartate aminotransferase from Escherichia coli in wild type (WT) as well as in a mutant form in which the highly conserved cis-proline at position 138 was replaced by a trans-alanine (P138A). Fluorescence analysis indicates that, although M* is an off-pathway intermediate in the folding of WT aspartate aminotransferase from E. coli, it seems to coincide with an on-pathway folding intermediate for the P138A mutant. Spectroscopic data, hydrogen/deuterium exchange, and limited proteolysis experiments demonstrated the occurrence of conformational differences between the two M*intermediates, with P138A-M* being conceivably more compact than WT-M*. Limited proteolysis data suggested that these conformational differences might be related to a different relative orientation of the small and large domains of the protein induced by the presence of the cis-proline residue at position 138. These differences between the two M* species indicated that in WT-M* Pro138 is in the cis conformation at this stage of the folding process. Moreover, hydrogen/deuterium exchange results showed the occurrence of few differences in the native N2 forms of WT and P138A, the spectroscopic features and crystallographic structures of which are almost superimposabl

    Design of a pH-responsive conductive nanocomposite based on MWCNTs stabilized in water by amphiphilic block copolymers

    Homogeneouswater dispersions ofmulti-walled carbon nanotubes (MWCNTs)were prepared by ultrasonication in the presence of an amphiphilic polystyrene-block-poly(acrylic acid) (PS-b-PAA) copolymer. The ability of PS-b-PAA to disperse and stabilizeMWCTNs was investigated by UV-vis, SEM and zeta potential. The results show that the addition of a styrene block to PAA enhances the dispersion efficiency of the graphitic filler compared to pure PAA, possibly due to the nanotube affinity with the polystyrene moiety. Notably, the dispersions show an evident pH-responsive behavior, being MWCNTs reaggregation promoted in basic environment.
